No i dont intend to rework the board. Since i am unable to use J501 Micro SD slot for sdmmc, can i use the these SDIO pins on the M.2 connector…

Are you trying to use a M.2 device that is based on SDIO? For example, a SDIO wifi card?

If not, then how do you expect to connect those pin on m.2 connector without rework the board?

If SDIO on M.2 is useable, then I plan to connect CYW54591 wifi card to M.2 slot on Nvidia Nano eval kit.

No, this is not supported on devkit as we didn’t test SDIO wifi card on that m.2 slot.
