I have Orin Nano developer kits booting from NVme with Jetpack 5.1.2.
- To upgrade them to Jetpack 6.0 should I again use SD cards or is there a way to do it without SD cards?
- Is there a way to update it within the device like it was possible to update from 5.1.1 to 5.1.2 or is a full flashing with SDK manager required?

You don’t need SDCards if you have a different storage device. You do need SDKManager to to a full reflash.
I want to add that there is QSPI memory on the module itself, and that this contains the equivalent of a BIOS plus boot content for SD card models. That content, as used in JetPack 5/L4T R35.x, is incompatible with JetPack 6/L4T R36.x DP. This is why (so far as I know) there is no upgrade path for that without flash.
