You can choose either sdcard or NVMe here.
There is actually a QSPI NOR-flash on Jetson itself to install bootloader. It is this bootloader that decides whether boot from sdcard or NVMe. So even without sdcard, this board can still use other medium to boot.
Keep the jumper throughout the flash. Once the flash is done, remove the jumper.
